Canine hemophilia and hemopoietic grafting.

چکیده
Two hemophilic beagles were given normal sibling. During observation 1200 A whole-body irradiation followed periods of 7 and 24 mo, there was no by a successful marrow graft from a evidence of factor VIII synthesis. ORTHOTOPIC TRANSPLANTATION of a normal liver into a hemophilic dog results in complete correction of the deficiency of factor VIII.”3 However, there are significant extrahepatic sources of factor VIII, since hepatectomized normal dogs bearing a transplanted liver from a hemophilic animal show factor VIII levels equivalent to that seen in the heterozygous state.2 3

Molecular cloning of a cDNA encoding canine factor IX.
Factor IX (F.IX) is a vitamin K-dependent plasma protein, a deficiency of which results in hemophilia B. A canine model of hemophilia B exists; attempts to use this model for gene transfer experiments or characterization of the hemophilic defect require elucidation of normal canine F.IX structure. We report the isolation and characterization of the coding region for canine F.IX cDNA. Canine F.I...
متن کاملComparison of Autogenic Costal Cartilage with Chitosan Scaffold in Canine Humeral Defect Healing
Objective- Current trends emphasize the acceleration of fracture healing on the ground that in doing so, the limitation of mobility and complications associated with recovery period are reduced. The present study aims to compare autogenic costal cartilage with Chitosan scaffold in canine humeral defect healing. Design- Experimental study Animal-15 adult male dogs Procedures-...
متن کاملSuccessful treatment of canine hemophilia by continuous expression of canine FVIIa.
Continuous expression of activated factor VII (FVIIa) via gene transfer is a potential therapeutic approach for hemophilia patients with or without inhibitory antibodies to human factor VIII (FVIII) or IX (FIX). Here, we investigate whether gene transfer of an engineered canine FVIIa (cFVIIa) transgene can affect hemostasis in a canine model of hemophilia, a good predictor of efficacy of hemoph...
متن کاملSystemic delivery of an adenoviral vector encoding canine factor VIII results in short-term phenotypic correction, inhibitor development, and biphasic liver toxicity in hemophilia A dogs.
Canine hemophilia A closely mimics the human disease and has been used previously in the development of factor VIII (FVIII) protein replacement products. FVIII-deficient dogs were studied to evaluate an in vivo gene therapy approach using an E1/E2a/E3-deficient adenoviral vector encoding canine FVIII. Results demonstrated a high level of expression of the canine protein and complete phenotypic ...
متن کاملSustained phenotypic correction of canine hemophilia A using an adeno-associated viral vector.
Gene therapy for hemophilia A requires efficient delivery of the factor VIII gene and sustained protein expression at circulating levels of at least 1% to 2% of normal. Adeno-associated viral type 2 (AAV2) vectors have a number of advantages over other viral vectors, including an excellent safety profile and persistent gene expression. However, a major disadvantage is their small packaging capa...
